Colorectal, breast, head and neck, cervical, urothelial and stomach cancers along with several lymphomas, pancreatic and skin cancers showed weak to moderate cytoplasmic and nuclear positivity. Remaining cancers were negative.
Several cases of stomach cancers showed moderate to strong cytoplasmic, membranous and nuclear staining while few cases of colorectal cancers displayed moderate positivity. Few cases of melanoma and prostate cancers showed moderate cytoplasmic immunoreactivity while several cases of breast cancers, few cases of skin cancers along with head and neck cancers showed additional nuclear staining. Remaining cancer tissues were weakly stained or negative.
Cancer tissues showed cytoplasmic immunoreactivity in most cases moderate. Several cases of breast cancer and a few cases of melanomas, cervical cancers and occasional cases of several other tumors types were strongly stained. Prostate cancer, carcinoid, thyroid and testis cancers were mainly negative.

This gene encodes an adaptor protein that is composed of two protein-protein interaction domains: a N-terminal PYRIN-PAAD-DAPIN domain (PYD) and a C-terminal caspase-recruitment domain (CARD). The PYD and CARD domains are members of the six-helix bundle death domain-fold superfamily that mediates assembly of large signaling complexes in the inflammatory and apoptotic signaling pathways via the activation of caspase. In normal cells, this protein is localized to the cytoplasm; however, in cells undergoing apoptosis, it forms ball-like aggregates near the nuclear periphery.
