A few skin and urothelial cancers displayed strong cytoplasmic immunoreactivity with a dot-like pattern. Some colorectal and pancreatic cancers were moderately stained. Most other malignant cells were negative.
Moderate to strong cytoplasmic positivity with a granular pattern was observed in a few cases of squamous cell carcinomas and urothelial cancers and in one case of basal cells carcinoma. Remaining malignant cells were negative.
Keratinized material of squamous cell carcinomas stained distinctly. Remaining malignant cells were negative.

It is initially synthesized as a polyprotein precursor, profilaggrin (consisting of multiple filaggrin units of 324 aa each), which is localized in keratohyalin granules, and is subsequently proteolytically processed into individual functional filaggrin molecules.
